  PHARMACOLOGICAL CLASSIFICATION Anti-Ulcer (Proton-Pump Inhibitor)

 VLANZOL belongs to a class of antisecretory compounds, the substituted benzimidazoles, that suppress gastric acid secretion by specific inhibition of the (H+, K+)-ATPase enzyme system at the secretory surface of the gastric parietal cell. Because this enzyme system is regarded as the acid (proton) pump within the parietal cell, Lansoprazole has been characterized as a gastric acid-pump inhibitor, in that it blocks the final step of acid production. This effect is dose-related and leads to inhibition of both basal and stimulated gastric acid secretion irrespective of the stimulus. Lansoprazole does not exhibit anticholinergic or histamine type-2 antagonist activity

 
 

PHARMACOKINETICS

VLANZOL is rapidly absorbed after oral administration, with peak plasma concentrations achieved about 1.5 hours after a dose by mouth. Bioavailability is reported to be 80% or more even with the first dose, although the drug must be given in an enteric-coated form since lansoprazole is unstable at acid pH. It is extensively metabolized in the liver primarily by the Cytochrome P450 isoenzyme CYP2C19 to form 5-hydroxyl-lansoprazole and by CYP3A to form lansoprazole sulfone. Metabolites are excreted primarily in faeces via the bile; only about 15 to 30% of a dose is excreted in urine. The plasma elimination half-life is around 1 to 2 hrs but the duration of action is much longer. Lansoprazole is about 97% bound to plasma protein. Clearance is decreased in elderly patients, and in liver disease. 

 
 

INDICATIONS

VLANZOL is indicated for the treatment of:

·          Active Duodenal Ulcer

·          Active Benign Gastric Ulcer

·          NSAID-Associated Gastric Ulcer

·          Gastroesophageal Reflux Disease (GERD)

·          Erosive Esophagitis

BEFORE YOU TAKE VLANZOL

Make sure you have told your doctor if you:·          are pregnant or planning to become pregnant ·          are breastfeeding
 
  HOW TO TAKE VLANZOL You must take your medicine as the doctor tells you. It is important to take VLANZOL Tablets at the right time. Your doctor will decide which is the best dose for you, depending on your illness.
  • VLANZOL is usually taken once a day.
  • Swallow each VLANZOL whole with water. Do not chew, crush, or split.
IF YOU MISS A DOSETake the missed dose as soon as you remember it. If it is almost time for your next dose, skip the dose you missed and take your next dose when you are meant to. Otherwise, take it as soon as you remember, and then go back to taking your doses as you would normally. If you are not sure whether to skip the dose, talk to your doctor or pharmacist. Do not take a double dose to make up for the dose that you missed. 

WHAT TO DO IF AN OVERDOSE IS TAKEN

In that case do not delay, ask your doctor immediately what to do or contact your nearest hospital emergency department. Do this even if there are no signs of discomfort or poisoning.  

TAKING OTHER MEDICINE

 Please tell your doctor if you are taking or have recently taken any other medicines, including medicines obtained without prescription, your doctor may wish to alter your dose of VLANZOL if you are taking any of the following:·          cyclosporine ·          digoxin ·          ketoconazole ·          warfarin   
 

SIDE EFFECTS

 Like other proton pump inhibitors, VLANZOL may cause serious allergic reactions. Symptoms of a serious allergic reaction include:
  • swelling of your face, eyelids, lips, tongue, or throat, and trouble swallowing
  • asthma (wheezing) or other breathing problems such as chest tightness or shortness of breath
  • Shock (loss of blood pressure and consciousness).
 
 
STORING
  • Store in a dry place at room temperature.
  • Keep all medicines out of the reach of children.
