PHARMACOLOGICAL CLASSIFICATION

  
  Antifungal
Fluconazole is a highly selective inhibitor of fungal cytochrome P-450 dependent enzyme lanosterol 14-α-demethylase. This enzyme functions to convert lanosterol to ergosterol. The subsequent loss of normal sterols correlates with the accumulation of 14-α-methyl sterols in fungi and may be responsible for the fungistatic activity of fluconazole.

PHARMACOKINETICS

Following oral dosing, fluconazole is almost completely absorbed within two hours. Bioavailability is not significantly affected by the absence of stomach acid. Concentrations measured in the urine, tears and skin are approximately 10 times the plasma concentration, while saliva, sputum and vaginal fluid concentrations are approximately equal to the plasma concentration, following a standard dose range of between 100 mg and 400 mg per day. The elimination half-life of fluconazole follows zero order kinetics and only 10% of elimination is due to metabolism, the remainder is excreted in urine and sweat.

INDICATIONS

ELFLUCO-150 is indicated for the treatment and prophylaxis of fungal infections:

1.        Vaginal candidiasis (vaginal yeast infections due to Candida). 2.        Oropharyngeal and esophageal candidiasis. 3.        Effective for the treatment of Candida urinary tract infections, peritonitis, and systemic Candida infections including candidemia, disseminated candidiasis, and pneumonia. 4.        Cryptococcal meningitis.

ELFLUCO-150 can be used first-line for the following indications:

Make sure you have told your doctor if you:

·          Have or have ever had liver disease. ·          Are pregnant, plan to become pregnant, or are breast-feeding.·           Have or have ever had cancer; acquired immunodeficiency syndrome (AIDS); ·           An irregular heartbeat; or heart, or kidney disease. 
 

HOW TO TAKE ELFLUCO-150  

 

You must take your medicine as the doctor tells you. It is important to take ELFLUCO-150 Tablets at the right time. Your doctor will decide which is the best dose for you, depending on your illness. Dosage varies with indication and between patient groups, ranging from: a two week course of 150 mg/day for vulvo vaginal candidiasis, to 150–300 mg once weekly for resistant skin infections or some prophylactic indications. 50–600 mg/day may be used for systemic or severe infections, and in urgent infections such as meningitis caused by yeast 800 mg/day have been used. Pediatric doses are measured at 6-12 mg/kg/d.

 IF YOU MISS A DOSE
Take the missed dose as soon as you remember it. If it is almost time for your next dose, skip the dose you missed and take your next dose when you are meant to. Otherwise, take it as soon as you remember, and then go back to taking your doses as you would normally. If you are not sure whether to skip the dose, talk to your doctor or pharmacist. Do not take a double dose to make up for the dose that you missed. 

WHAT TO DO IF AN OVERDOSE IS TAKEN

In that case do not delay, ask your doctor immediately what to do or contact your nearest hospital emergency department. Do this even if there are no signs of discomfort or poisoning. Symptoms of overdose may include confusion, extreme sweating, blurred vision, or unusually fast heartbeat.

TAKING OTHER MEDICINE

 

Please tell your doctor if you are taking or have recently taken any other medicines, including medicines obtained without prescription, your doctor may wish to alter your dose of ELFLUCO-150 if you are taking:

·          Anticoagulants (blood thinners) such as warfarin

·          Astemizole·          Benzodiazepines·          Diuretics·          Erythromycin·          Isoniazid·          Moxifloxacin

·          oral contraceptives

SIDE EFFECTS

  
Common side effects of ELFLUCO-150 include:
 
  • headache
  • diarrhea
  • nausea or upset stomach
  • dizziness
  • stomach pain
  • changes in the way food tastes
Some side effects can be serious. If you experience any of the following symptoms, call your doctor immediately:
 
  • upset stomach
  • extreme tiredness
  • unusual bruising or bleeding
  • lack of energy
  • loss of appetite
  • pain in the upper right part of the stomach
  • yellowing of the skin or eyes
  • flu-like symptoms
  • dark urine
  • pale stools
  • seizures
STORING
 
 Keep all medicines out of the reach of children ·          Store it at room temperature
