PHARMACOLOGICAL CLASSIFICATION   

 MECHANISM OF ACTION
Ibuprofen is an NSAID which is believed to work through inhibition of cyclooxygenase (COX), thus inhibiting prostaglandin synthesis. There are at least 2 variants of cyclooxygenase (COX-1 and COX-2). Ibuprofen inhibits both COX-1 and COX-2. It appears that its analgesic, antipyretic, and anti-inflammatory activity are achieved principally through COX-1 inhibition; whereas COX-2 inhibition is responsible for its unwanted effects on platelet aggregation and the GI mucosa. 

PHARMACOKINETICS

Ibuprofen is absorbed from the gastrointestinal tract and peak plasma concentrations occur about 1 to 2 hours after ingestion. Ibuprofen is 90 to 99% bound to plasma proteins and has a plasma half-life of about 2 hours. It is rapidly excreted in the urine mainly as metabolites and their conjugates. About 1% is excreted in urine as unchanged ibuprofen and about 14% as conjugated ibuprofen. There appears to be little if any distribution into breast milk.         

 In Children Ibuprofen Oral Suspension is indicated:For the reduction of fever in patients aged 6 months and older.For the relief of signs and symptoms of juvenile arthritis.In Adults Ibuprofen Oral Suspension is indicated:For the treatment of rheumatic diseases such as: Rheumatoid arthritis, Osteoarthritis, Ankylosing spondylitis, Juvenile rheumatoid arthritisFor the relief of mild to moderate pain especially when anti- inflammatory actions may also be desired e.g., following dental or obstetric surgery, and for relief of musculoskeletal pain due to soft tissue athletic injuries (strains or sprains).For the relief of pain and inflammation of acute gouty arthritis.
In the treatment of painful nonrheumatic inflammatory conditions such as athletic injuries, bursitis, capsulitis, synovitis, tendinitis or tenosynovitis.
For reduction of fever.
For the relief of pain associated with primary dysmenorrhoea

BEFORE YOU TAKE IBUVIL

Do not take IBUVIL if:You have ever had or are suffering from stomach ulcers.You are allergic to ibuprofen, any of the ingredients, aspirin or any other Non Steroidal Anti-Inflammatory Drugs (NSAIDs).(an allergic reaction may be recognised as shortness of breath,runny nose, skin rash or itching).Talk to your doctor before taking if:You suffer from asthma.You are pregnant or breastfeeding.You are elderly.You have heart, liver or kidney problems.You are taking regular medication, especially drugs for high blood pressure (eg. water tablets), heart disease or anticoagulants (to prevent blood clotting).you are allergic to or taking any other painkillers. HOW TO TAKE IBUVIL

  • Take Ibuprofen exactly as directed by your doctor.
  • Always read the manufacturers information leaflet, if possible, before beginning treatment.
  • Take the tablet and oral liquid form of Ibuprofen with or immediately after food.
  • Ibuprofen is for you. Never give it to others even if their condition appears to be the same as yours.

IF YOU MISS ADOSEGive the next dose when needed. Do not give a double dose.

WHAT TO DO IF AN OVERDOSE IS TAKEN

In that case do not delay, ask your doctor immediately what to do or contact your nearest hospital emergency department. TAKING OTHER MEDICINEIf you are on low dose aspirin - medicines for high blood pressure and water tablets (diuretics)- medicines for thinning the blood (anti-coagulants)

- Methotrexate
- Lithium
- Zidovudine  
  

SIDE EFFECTS

Side effects that may occur include:

· Stomach pain, nausea, indigestion and occasionally stomach ulcer.

· Occasionally bleeding of the stomach may occur (signs of this are vomiting ofblood and black stools), if you notice either of these symptoms stop taking theproduct and see your doctor immediately.

· Itchy or swollen skin or skin rashes, and very rarely skin peeling.

· Blood disorders which can cause bruising and sometimes abnormal bleeding.Very rarely liver or kidney problems, headache, dizziness and hearing disturbances.If you experience any of the following, stop taking the medicine immediately and tellyour doctor:Unexplained wheezing, shortness of breath, skin rash, itching, bruising, or facialswelling. 

STORING  

  • Keep all medicines out of the reach of children.
  • Store in a cool dry place, away from direct heat and light.

Never keep out of date or unwanted medicines. Discard them safely out of the reach of children.
